Gamepad is required to play. TwinklePop just wants everyone to be happy! Use the right trigger to share your happiness with the other creatures. Make sure not to get too close or they'll reject you.
The game didn't seem to recognize my Logitech RumblePad 2 game controller. I found that the keyboard arrow keys would change TwinklePop's direction of facing (but not move him) and pressing space would send a rainbow beam out always to the right. Hit a few of the cats with the rainbow but never got any points for them. TwinklePop reminded me of the ghost from Bubble Ghost back in the day, so some nice nostalgia factor there.
